Daypro had a higher reporting rate of Stevens-Johnson Syndrome (SJS) and Topical Epidermal Necrolysis than any other NSAID in its first year on the market, including four deaths. Daypro caused more deaths from skin reactions in the five year period from 1993 to 1997 than any other prescription NSAID on the market.
History
Daypro was approved by the FDA in 1992. It took more than ten years and three rejections for Searle to get the drug approved. The problems with approval included the FDA's opinion that oxaprozin was no more effective than other arthritis drugs already available, and that oxaprozin had a concerning number of adverse reactions and effects.
Females seem to be in particular danger for SJS/TEN reactions from Daypro.Approximately 80% of the reported adverse reactions were from women. This trend did show up in the clinical trials of the drug, but the drug was approved anyway. Low body weight is another indicator of increased risk for toxic reactions to Daypro. This may be related to the fact that Daypro has a significantly long half-life. A drug's half-life means the time it takes for the drug's concentration in the blood to be reduced by half. A longer half-life means the drug stays in the blood stream for a longer period of time and at a heavier concentration. That can be considered a good thing for the therapeutic aspects of a drug, but is a negative quality for toxic reactions.
Adverse drug reactions are the 4th leading cause of death in North America.Daypro/Oxaprozin has caused serious adverse drug reactions and side effects from Stevens-Johnson Syndrome and TEN, including death, blindness and disfigurement.
